Sensitivity Differences between Several Primers for Detection of Mycobacterium tuberculosis by PCR Method

Abstract
Recently research efforts and clinical trial studies have been focused on finding new methods for the rapid assessment of mycobacteria from various clinical samples. Severel methods have been developed which shorten detection period. Among them,
polymerase chain reaction(PCR) method is thought to be the fastest and sensitive method, But standarsization of this method is not yet accomplished. Here we tried to compare the sensitivity of some primer pairs for the detection of M.
tuberculosis.
We tested three primer located within the IS6110 sequence of M. tuberculosis. PCRs were performed in various conditions with these primer pairs and the results were shown on electrophoresed agarose gel stained with ethidium bromide. All three
primer
pairs showed clear amplified DNA bands. Among them, 123 bp DNA had shown some difficulty on electrophoresis and viewing on UV light. Primer pair for 245bp DNA sequence showed slightly more sensitivity than primer pair for 359 bp DNA. Primer pair
for 245
bp DNA in IS6110 seems to be the most useful for its handling convenience and sensitivity.
